Doxorubicin HCl
Indications/Uses
Solid tumours, haemoblastoses & lymphomas, intravesical application for superficial, non-invasive urinary bladder tumours after transurethral resection, relapse prevention. Various types of neoplasia.
Dosage/Direction for Use
60-75 mg/m2 BSA as single IV infusion at 3 wk intervals. Cumulative dosage should not exceed 500 mg/m2. Patient w/ preexisting cardiac disease or previously receiving cardiac irradiation Max 400 mg/m2. Superficial bladder cancer & bladder carcinoma in-situ 50 mg in 50 mL physiological NaCl through sterile catheter.
Contraindications
Myelosuppression, cardiopathy, buccal ulceration. History of receiving cardiotoxic agents, cyclophosphamides. UTI, cystitis. Urethral stenosis. Pregnancy & lactation.
Special Precautions
Check liver functions, cardiac parameters & blood counts before & during treatment. Monitor haematological parameters. Regularly monitor BUN & sufficient fluid intake. Superficial bladder cancer & carcinoma. Hepatic impairment. May impair ability to drive or operate machinery. Childn. Elderly.
Adverse Reactions
Bone marrow depression. Cardiotoxicity, irreversible CHF, reversible alopecia, hyperpigmentation of nailbeds, dermal creases, onycholysis, nausea, vomiting, anorexia, stomatitis, esophagitis, ulceration & necrosis of the colon, diarrhea, phlebosclerosis, severe cellulitis, vesication, tissue necrosis, fever, chills, urticaria, anaphylaxis, conjunctivitis, lacrimation, mucositis, hyperuricaemia.
Drug Interactions
Verapamil, cyclophosphamide, methotrexate, cyclosporine, streptozocin. CYP450 inhibitors.
