Pharmacology: An oral dosage form for treatment of erectile dysfunction. Its active ingredient, sildenafil, inhibits cGMP specific phosphodiesterase type-5 (PDE5) enhancing the effect of nitric oxide (NO) in the corpus cavernosum resulting in smooth muscle relaxation and inflow of blood to corpus cavernosum, hence penile erection maintains for some time.
Erectile dysfunction in males.
Adults: One tablet (50 mg) as a single dose 1 hour before sexual activity. Maximum Dose: Once per day. Alternatively, may be taken ½ to 4 hours before sexual activity. Depending on the effectiveness, the dose may be increased to a maximum of 2 tablets (100 mg).
Elderly: Initiate therapy at a lower dose.
Concurrent use with organic nitrates (e.g. nitroglycerin) may severely lower blood pressure hence is not recommended. It is also not recommended for use in patients hypersensitive to sildenafil and for use in women.
Caution is required in patients with hepatic or severe renal impairment and dosage reduction may be necessary. Care is also needed in patients with anatomical deformation of penis or hematological disorders. In case of prolonged erection for >4 hours, sudden visual or hearing loss, immediate medical attention is advised.
Usually moderate and transient such as headache, dizziness, flushing, abnormal vision (blurred vision and sensitive to light), indigestion and nasal congestion.
Do not take together with other drugs indicated for erectile dysfunction. When administered concurrently with cimetidine, delavirdine, erythromycin, itraconazole, ketoconazole, a reduction in dosage should be considered.
Store at temperature not more than 30°C.
