Indications/Uses
Schizophrenia in adults & adolescents 15-17 yr.
Dosage/Direction for Use
Schizophrenia Adult Initially 40 mg once daily. Dose range: 40-160 mg/day. Max: 160 mg/day. Adolescent 15-17 yr Initially 40 mg once daily. Dose range: 40-80 mg/day. Max: 80 mg/day.
Administration
Should be taken with food.
Contraindications
Hypersensitivity. Concomitant use w/ strong CYP3A4 inhibitors & inducers.
Special Precautions
Increased mortality & higher incidence of cerebrovascular adverse reactions in elderly patients w/ dementia-related psychosis. Neuroleptic malignant syndrome; tardive dyskinesia; metabolic changes (eg, hyperglycemia, dyslipidemia, body wt gain); hyperprolactinemia; leukopenia, neutropenia & agranulocytosis; orthostatic hypotension & syncope; elevation in core body temp; esophageal dysmotility & aspiration. Use cautiously in patients w/ a history of seizures or w/ conditions that lower the seizure threshold; patients at risk for aspiration pneumonia. Increased sensitivity to antipsychotic medication in patients w/ Parkinson's disease or dementia w/ Lewy bodies. Possibility of a suicide. Potential to impair judgment, thinking or motor skills. Observe carefully for signs of misuse or abuse. Moderate or severe renal or hepatic impairment. Pregnancy & lactation. Childn. Elderly.
Adverse Reactions
Somnolence, akathisia, extrapyrimidal symptoms, nausea.
Drug Interactions
Do not use concomitantly w/ strong CYP3A4 inhibitors (eg, ketoconazole, clarithromycin, ritonavir, voriconazole, mibefradil) or strong CYP3A4 inducers (eg, rifampin, avasimibe, St. John's wort, phenytoin, carbamazepine); grapefruit & grapefruit juice. Reduce dose to ½ when used concomitantly w/ moderate CYP3A4 inhibitors (eg, diltiazem, atazanavir, erythromycin, fluconazole, verapamil). Increasing the dose may be necessary when concomitantly used w/ moderate CYP3A4 inducer after chronic treatment.
