Losartan K 50 mg, hydrochlorothiazide 12.5 mg
Indications/Uses
Treatment of essential HTN in patients whose BP is not adequately controlled on losartan or hydrochlorothiazide alone.
Dosage/Direction for Use
1 tab once daily. May be increased up to max 100/25 mg once daily.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity to losartan K, hydrochlorothiazide, or other sulfonamide-derived drugs. Anuria.
Special Precautions
Pregnancy & lactation. Losartan Renal artery stenosis. Renal & hepatic impairment. Correct vol depletion prior therapy. Hyperkalaemia. Perform periodic determination of serum electrolytes. Progressive liver disease. Hydrochlorothiazide W/ or w/o history of allergy or bronchial asthma. Exacerbation or activation of SLE. Glucose tolerance. Hyperglycemia. Hyperuricemia. Post sympathectomy patients. Hypomagnesemia. Increased cholesterol & triglyceride levels.
Adverse Reactions
Losartan Dizziness, headache, dose-related orthostatic hypotension, hyperkalaemia, myalgia, arthralgia, renal function impairment, cough, resp tract disorders, back pain, GI disturbances, fatigue, neutropenia. Hydrochlorothiazide Weakness, pancreatitis, jaundice, sialadenitis, cramping, gastric irritation, aplastic anemia, agranulocytosis, leukopenia, hemolytic anemia, thrombocytopenia, purpura, phostosensitivity, urticaria, vasculitis, cutaneous vasculitis, fever, pneumonitis, pulmonary edema, hyperglycemia, glycosuria, hyperuricemia, muscle spasm, restlessness, renal failure, interstitial nephritis, Stevens-Johnson syndrome, toxic epidermal necrolysis, transient blurred vision, xanthopsia.
Drug Interactions
Losartan Decreased conc w/ rifampin. Increased conc w/ fluconazole. Increased serum K w/ K-sparing diuretics, K supplements, K-containing salt substitutes. Blunted antihypertensive effect w/ NSAIDs. Hydrochlorothiazide Potentiation of orthostatic hypotension w/ alcohol, barbiturates or narcotics. Oral antidiabetic agents, insulin. Reduced absorption w/ cholestyramine & colestipol resins. Intensified electrolyte depletion w/ corticosteroids, adrenocorticotropic hormone. Decreased response to pressor amines. Increased responsiveness w/ skeletal muscle relaxants, tubocurarine. Increased lithium toxicity. Reduced effects w/ NSAIDs including selective Cox-2 inhibitors.
