Gliclazide
Indications/Uses
DM of the maturity onset type, which cannot be controlled by diet alone.
Dosage/Direction for Use
Initially, 40 mg daily, may increase up to 320 mg daily. Doses >160 mg should be taken in 2 divided doses.
Administration
Should be taken with food.
Contraindications
Diabetes complicated by acidosis, ketosis or coma. History of repeated episodes of ketoacidosis or coma. Juvenile onset diabetes & unstable or brittle diabetes. Severe renal & hepatic insufficiency. Pregnancy & lactation.
Special Precautions
Acute complications eg, severe trauma, fever, infection or surgery.
Adverse Reactions
Hypoglycaemia, GI disturbances, dermatological reactions, biochemical abnormalities. Headache, slight disulfiram-like reactions, lassitude.
Drug Interactions
Thiazides, barbiturates, glucocorticoids & oestrogens. Insulin, biguanides, sulfonamides, oxyphenbutazone, phenylbutazone, clofibrate, salicylates, coumarin derivatives, chloramphenicol, MAOIs, β-blockers, cimetidine, ethanol. Alcohol.
