Indications/Uses
Dosage/Direction for Use
Benign gastric & duodenal ulcer 20 mg once daily for 4 wk (duodenal ulcer) or 8 wk (gastric ulcer). May be increased to 40 mg once daily in severe or recurrent cases. Recurrent duodenal ulcer 20 mg once daily up to 1 yr. Prevention of duodenal ulcer relapse 20-40 mg daily. GERD 20-40 mg once daily for 4-8 wk. Zollinger-Ellison syndrome Initially 60 mg once daily (range 20-120 mg daily). Doses >80 mg should be divided & given bd. H. pylori eradication Dual therapy: 40 mg daily w/ amoxicillin 750 mg bd or clarithromycin 500 mg tds for 2 wk. Triple therapy: 20 mg bd or 40 mg once daily w/ amoxicillin 500 mg tds & metronidazole 400 mg tds for 7 days, OR 20 mg bd or 40 mg once daily w/ clarithromycin 250 mg bd & metronidazole 400 mg (or tinidazole 500 mg) bd for 7 days, OR 20 mg bd or 40 mg once daily w/ amoxicillin 1 g bd & clarithromycin 500 mg bd for 7 days.
Administration
Should be taken with food: Take immediately before a meal.
Contraindications
Special Precautions
Increased risk of bacteria-associated diarrhoea. Hypomagnesemia on long-term use. Exclude malignancy if gastric ulcer is suspected. Chronic hepatic disease. Pregnancy & lactation. Childn. Elderly.
Adverse Reactions
Headache, diarrhoea, constipation, nausea, vomiting, flatulence, abdominal pain. In isolated cases or rarely, hepatitis w/ or w/o jaundice, hepatic failure.
Drug Interactions
Reduced absorption of ampicillin esters, Fe salts, ketoconazole. Prevented degradation of acid-labile drugs. Increased plasma conc w/ clarithromycin, of digoxin. Decreased hepatic metabolism of diazepam, phenytoin, warfarin. Increased leukopenic &/or thrombocytopenic effects w/ bone marrow depressants. Reduced pharmacological activity of clopidogrel.
