Indications/Uses
2nd or 3rd line treatment of type 2 DM as monotherapy in adults inadequately controlled by diet & exercise for whom metformin is inappropriate; as dual oral therapy in combination w/ metformin in adults w/ insufficient glycaemic control despite maximal tolerated dose; as triple oral therapy in combination w/ metformin & sulphonylurea in adults w/ insufficient glycaemic control despite dual oral therapy.
Dosage/Direction for Use
Initially 15 mg or 30 mg once daily. Dose may be increased in increments of up to 45 mg once daily.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. History of/or cardiac failure & bladder cancer. Hepatic impairment. Diabetic ketoacidosis. Macroscopic haematuria.
Special Precautions
Fluid retention & cardiac failure. Bladder cancer. Periodic monitoring of liver function. Wt gain. Reduced mean haemoglobin & haematocrit, consistent w/ haemodilution. Hypoglycaemia. Decreased visual acuity. Increased incidence of bone fractures in women. Resumption of ovulation. Glucose intolerance, Lapp lactase deficiency, glucose-galactose malabsorption. May affect ability to drive or operate machinery. Pregnancy & lactation. Childn & adolescent <18 yr.
Adverse Reactions
Hypoglycaemia, oedema, upper resp infection, bronchitis, anaemia, hypoasthesia, headache, dizziness, visual disturbance, heart failure, dyspnoea, bone fracture, arthralgia, back pain, haematuria, erectile dysfunction, increased wt.
Drug Interactions
Increased/decreased conc w/ gemfibrozil or rifampicin.
