Sildenafil citrate
Pulmonary arterial HTN in adult ≥18 yr.
Dosage/Direction for Use
May be taken with or without food.
Hypersensitivity. Concomitant use w/ nitrates in any form (oral, SL, skin patch or by inhalation); nitroglycerin; isosorbide mononitrate & isosorbide dinitrate.
Special Precautions
Heart or blood vessel diseases; temporary, decreased, or permanent loss of vision in 1 or both eyes including Non-Arteritic Anterior Ischemic Optic Neuropathy (NAION). Heart problems (irregular heartbeats, angina, chest pain, recent heart attack); high or low BP; kidney or liver problems; blood problems including sickle cell anemia or leukemia; deformed penis, Peyronie's disease, erection lasting >4 hr; stomach ulcers or any types of bleeding problems; retinitis pigmentosa; concomitant use of α-blocker, nitrates or any other medicines. Risk of serious skin reactions (eg, Stevens-Johnson Syndrome (SJS), toxic epidermal necrolysis, erythema multiforme). Do not operate a motor vehicle or any heavy machinery in case of visual disturbances. Pregnancy & lactation. Not indicated for use in childn <18 yr.
Adverse Reactions
Headache, facial flushing, indigestion, back pain, diarrhoea, nausea, limb pain; sudden decrease or loss of hearing; flu-like symptoms, difficulty sleeping, effects on vision, cough, nosebleed, muscle aches, increased temp, seizure, seizure recurrence or transient global amnesia (temporary memory loss).
Drug Interactions
Use of organic nitrates in any form is contraindicated. Potential interaction w/ α-blockers, bosentan; ketoconazole or itraconazole; ritonavir or certain HIV drugs; cimetidine; erythromycin. Grapefruit juice may affect sildenafil blood levels.
