Indications/Uses
Adults w/ type 2 DM to improve glycaemic control as monotherapy when diet & exercise alone do not provide adequate glycaemic control in patients for whom the use of metformin is considered inappropriate due to intolerance or contraindications; or as add-on therapy in combination w/ other glucose-lowering medicinal products including insulin, when these, together w/ diet & exercise, do not provide adequate glycaemic control.
Dosage/Direction for Use
SC Adult Monotherapy: 0.75 mg once wkly. Add-on therapy: 1.5 mg once wkly. Elderly ≥75 yr Initially 0.75 mg once wkly.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Do not use in patients w/ type 1 DM or for treatment of diabetic ketoacidosis. Not a substitute for insulin. Potential risk of dehydration in relation to GI side effects; avoid fluid depletion. Not recommended in patients w/ severe GI disease eg, severe gastroparesis. Risk of acute pancreatitis. Increased risk of hypoglycaemia in combination w/ sulphonylurea or insulin. Patients w/ CHF. Avoid driving & using machines in the presence of hypoglycaemia. Not recommended in patients w/ end stage renal disease (<15 mL/min/1.73 m2). Not recommended during pregnancy. Should not be used during breast-feeding. Childn <18 yr.
Adverse Reactions
Hypoglycaemia; nausea, diarrhoea, vomiting, abdominal pain. Decreased appetite, dyspepsia, constipation, flatulence, abdominal distention, GERD, eructation; fatigue; sinus tachycardia, 1st degree AV block.
Drug Interactions
May impact absorption rate of concomitantly administered oral medicinal products. May slightly increase drug exposure of some PR formulations. Increased exposure w/ sitagliptin leading to enhanced effects on blood glucose levels.
