Donepezil HCl
Indications/Uses
Symptomatic treatment of mild to moderately severe Alzheimer's dementia.
Dosage/Direction for Use
Adult/elderly Initially 5 mg/day in the evening for at least 1 mth. Max: 10 mg/day.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity to donepezil & piperidine derivatives.
Special Precautions
Severe Alzheimer's dementia, other types of dementia & memory impairment. Heart block, long sinus pauses, increased risk of ulcers, bladder outflow obstruction, exacerbation or induction of extrapyramidal symptoms, history of asthma, or obstructive pulmonary disease. Galactose intolerance, Lapp lactase deficiency, glucose-galactose malabsorption. Severe hepatic impairment. Pregnancy & lactation.
Adverse Reactions
Diarrhoea, muscle cramps, fatigue, nausea, vomiting, insomnia, headache.
Drug Interactions
Increased conc w/ ketoconazole, quinidine, CYP3A4 (itraconazole, erythromycin) & 2D6 inhibitors (fluoxetine). Decreased conc w/ rifampicin, phenytoin, carbamazepine & alcohol. Synergistic activity w/ succinylcholine, other neuromuscular blockers, cholinergic agonists, β-blockers.
