Indications/Uses
Addisonian pernicious anemia. Prophylaxis & treatment of other macrocytic anaemia associated w/ vit B12 deficiency. Schilling test.
Dosage/Direction for Use
IM Addisonian pernicious anaemia & other macrocytic anaemia w/o neurological involvement Initially 250-1000 mcg on alternate days for 1-2 wk, then 250 mcg wkly until blood count is normal. Maintenance: 1000 mcg mthly; w/ neurological complications 1000 mcg on alternate days until improvement occur. Prophylaxis of macrocytic anaemia associated w/ vit B12 deficiency from gastroectomy, some malabsorption syndromes & strict vegetarism 250-1000 mcg mthly. Schilling test 1000 mcg.
Contraindications
Hypersensitivity. Childn <3 yr.
Special Precautions
Not for toxic amblyopias, megaloblastic anaemia of pregnancy. Regular monitoring of the blood & plasma K. Dosage >10 mcg daily may produce haematological response in folate deficiency patients. Cardiac arrhythmias secondary to hypokalaemia.
Adverse Reactions
Itching, exanthema, chills, fever, hot flushes, nausea & dizziness, anaphylactic shock; acneiform & bullous eruptions.
Drug Interactions
Decreased patient's response w/ chloramphenicol.
