Oxaliplatin
Indications/Uses
Metastatic colorectal cancer.
Dosage/Direction for Use
Adult Recommended dose: 85 mg/m2 at a rate of 2-6 hr IV infusion prior to fluoropyrimidine, repeated every 2 wk.
Contraindications
Severe myelosuppression, impaired renal function, allergic reactions; peripheral sensory neuropathy w/ functional impairment prior to 1st course. Pregnancy & lactation. Childn.
Special Precautions
Anaphylactic-like reactions may occur. History of allergic reactions to platinum compounds. Discontinue when oxaliplatin extravasation occurs. Perform neurological exam before initiation of each administration & periodically thereafter. Severe diarrhea/emesis. If hematological toxicity occurs, administration of the next course of therapy should be postponed until values returned to normal. OC should be taken prior to initiation of treatment.
Adverse Reactions
Primary sensory peripheral neuropathy, pharyngolaryngeal dysaesthesia, jaw spasm, abnormal tongue sensation, feeling of chest pressure. Anaemia, neutropenia, thrombocytopenia. diarrhoea, nausea, vomiting, stomatitis, anorexia, abdominal pain, mucositis, dehydration, ileus, intestinal obstruction, hypokalemia, metabolic acidosis, constipation, elevation of transaminases & alkaline phosphatase activities. Back pain, arthralgia. Skin rash, conjunctivitis, rhinitis. Altered renal function. Infection fever, febrile neutropenia. Alopecia, rash.
