Aciclovir
Dosage/Direction for Use
IV infusion Herpes simplex (except herpes encephalitis) Adult 5 mg/kg infused over 1 hr 8 hrly for 7 days. Childn 3 mth-12 yr 250 mg/m2 infused over 1 hr 8 hrly for 7 days. Herpes simplex encephalitis Adult 10 mg/kg infused over 1 hr 8 hrly for 10 days. Childn 6 mth-12 yr 500 mg/m2 infused over 1 hr 8 hrly for 10 days. Severe initial episodes of herpes simplex genitalis Adult 5 mg/kg infused over 1 hr 8 hrly for 5 days. Immune-compromised patients w/ varicella zoster infection Adult 10 mg/kg infused over 1 hr 8 hrly for 7 days. Childn 500 mg/m2 infused over 1 hr 8 hrly for 7 days.
Contraindications
Hypersensitivity to aciclovir or valaciclovir.
Special Precautions
Dehydrated or renal impairment. Pregnancy & lactation.
Adverse Reactions
Nausea, vomiting, increased liver-related enzymes, rash, fever, decreased hematological indices & severe local inflammatory reactions when inadvertently infused into extravascular tissues. Occasionally, rapid increases in blood urea & creatinine levels.
Drug Interactions
Increased plasma conc w/ probenecid & cimetidine.
