Lamivudine
Indications/Uses
In combination w/ other antiretroviral for treatment of HIV infected adult & childn.
Dosage/Direction for Use
Tab Adult, adolescent & childn (at least 25 kg) 300 mg daily or 150 mg bd. Childn ≥20 kg to <25 kg ½ tab in the morning & 1 tab in the evening. 14 to <20 kg ½ tab bd or 1 tab once daily. Oral soln Adult & adolescent (at least 25 kg) 30 mL once daily or 15 ml bd. Childn >3 mth (<25 kg) 4 mg/kg bd. Max: 300 mg daily.
Administration
May be taken with or without food.
Contraindications
Special Precautions
Not recommended for use as monotherapy. Discontinue use if pancreatitis occurs. Lactic acidosis/severe hepatomegaly w/ steatosis. Immune reconstitution syndrome. Manage serum lipids & blood glucose. Periodic monitoring of liver function tests & HBV replication markers upon discontinuation in patients w/ HIV & HBV co-infection. Moderate to severe renal impairment, risk factor for liver disease. Pregnancy. Not to be used during lactation. Elderly. Oral soln: Diabetic patients. Monitor HIV-1 viral load w/ sorbitol-containing drugs.
Adverse Reactions
Hyperlactataemia; headache; nausea, vomiting, upper abdominal pain, diarrhoea; rash, alopecia; arthralgia, muscle disorders; fatigue, malaise, fever.
Drug Interactions
Decreased AUC & Cmax w/ sorbitol. Increased Cmax of zidovudine. Increased exposure w/ trimethoprim/sulphamethoxazole. Inhibited intracellular phosphorylation of emtricitabine.
