Contents
Per 625 mg FC tab Amoxicillin trihydrate 500 mg, K clavulanate 125 mg. Per 1 g FC tab Amoxicillin trihydrate 875 mg, K clavulanate 125 mg. Per 228 mg/5 mL oral susp Amoxicillin trihydrate 200 mg, K clavulanate 28.5 mg. Per 457 mg/5 mL oral susp Amoxicillin trihydrate 400 mg, K clavulanate 57 mg. Per 600 mg inj Amoxicillin Na 600 mg, K clavulanate 100 mg. Per 1.2 g inj Amoxicillin Na 1 g, K clavulanate 200 mg
Indications/Uses
Short-term treatment of upper & lower resp tract, skin & soft tissue, GUT infections. Tab & inj Short-term treatment of bone & joint & other infections. Tab & oral susp Dental infections. Inj Surgical prophylaxis against infections.
Dosage/Direction for Use
Tab Adult & childn >12 yr Mild to moderate infections 625 mg bd. Severe infections 1 g bd. Adult CrCl >30 mL/min 625 mg bd or 1 g bd. CrCl 10-30 mL/min 625 mg bd, CrCl <10 mL/min Not >625 mg every 24 hr. 228 mg/5 mL oral susp Mild to moderate infections Childn 7-12 yr (22-40 kg) 10 mL bd, 2-6 yr (13-21 kg) 5 mL bd. Severe infections 2-6 yr (13-21 kg) 10 mL bd. 457 mg/5 mL oral susp Mild to moderate infections 7-12 yr (22-40 kg) 5 mL bd, 2-6 yr (13-21 kg) 2.5 mL bd. Severe infections 7-12 yr 10 mL bd, 2-6 yr (13-21 kg) 5 mL bd. Inj Adult, childn >12 yr & CrCl >30 mL/min 1.2 g 8 hrly, may be increased to 6 hrly. Childn 3 mth-12 yr 30 mg/kg 8 hrly, may be increased to 6 hrly, 0-3 mth 30 mg/kg every 12 hr, may be increased to every 8 hr. Surgical prophylaxis 1.2 g IV at anesth induction up to 3-4 doses in 24-hr period. CrCl 10-30 mL/min 1.2 g IV stat, followed by 600 mg IV 12 hrly, CrCl <10 mL/min 1.2 g IV stat, followed by 600 mg IV 24 hrly; additional 600 mg IV may need to be given during dialysis & at end of dialysis.
Administration
May be taken with or without food: Best taken at the start of meals for better absorption & to reduce GI discomfort. Tab: May be broken in half & swallowed whole w/o chewing.
Contraindications
Hypersensitivity to β-lactams eg, penicillins, cephalosporins. History of penicillin-associated jaundice/hepatic dysfunction.
Special Precautions
Discontinue use if allergic reaction occurs. Avoid use if infectious mononucleosis is suspected. Overgrowth of non-susceptible organisms. Pseudomembraneous colitis, cholestatic jaundice. Monitor prothrombin time in patients receiving anticoagulants. Maintain adequate fluid intake & urinary output during high doses. Hepatic & renal impairment. Avoid use during 1st trimester of pregnancy. Tab: Not recommended in childn ≤12 yr. Oral susp: Phenylketonuria. Not recommended in renal impairment. Not recommended in childn w/ GFR <30 mL/min & infants w/ immature renal function. Inj: Not for IM inj. Na restricted diet. False +ve Coombs' test.
Adverse Reactions
Diarrhoea. Mucocutaneous candidiasis; nausea, vomiting.
Drug Interactions
Decreased renal tubular secretion by probenecid. Increased likelihood of allergic skin reactions w/ allopurinol. Lowered oestrogen reabsorption & reduced efficacy of combined OCs. Increased INR w/ acenocoumarol or warfarin. Reduced pre-dose conc of mycophenolic acid.
