Zuclopenthixol (Drops: diHCl; Acuphase inj: acetate; Depot inj: decanoate)
Dosage/Direction for Use
Clopixol oral drops Acute schizophrenia & other acute psychoses, severe acute state of agitation & mania 10-50 mg daily, max: 75 mg or more. Chronic schizophrenia & other chronic psychoses 20-40 mg daily. Agitation in the mentally retarded 6-20 mg daily, increased if necessary to 25-40 mg daily. Agitation & confusion in senile patient 2-6 mg daily, increased if necessary to 10-20 mg daily. Clopixol-Acuphase inj Initial treatment of acute psychoses, including mania & exacerbation of chronic psychoses 50-150 mg IM repeated if necessary, preferably w/ time interval of 2-3 days. In a few patients an additional inj may be needed 24-48 hr after the 1st inj. Clopixol Depot inj Maintenance treatment of schizophrenia & other psychoses 200-400 mg IM every 2-4 wk.
Administration
May be taken with or without food.
Contraindications
Comatose states, acute alcohol, barbiturate & opiate intoxication. Pregnancy.
Special Precautions
Patients on long-term therapy should be monitored carefully. Advanced hepatic & CV disease, organic brain syndrome & convulsive disorders. May affect ability to drive or operate machinery.
Adverse Reactions
Extrapyramidal syndrome; drowsiness, dry mouth, urinary retention, disturbed accommodation, tachycardia, postural hypotension, dizziness. Tardive dyskinesia.
Drug Interactions
Alcohol, barbiturates, CNS depressants, guanethidine, metoclopramide.
