Contents
Atovaquone 250 mg, proguanil HCl 100 mg
Indications/Uses
Prophylaxis & treatment of Plasmodium falciparum malaria especially in areas where the pathogen may be resistant to other antimalarials.
Dosage/Direction for Use
Prophylaxis Start 1-2 days before entering malaria-endemic area & continue daily until 7 days after leaving the area. Adult 1 tab daily. Treatment Adult & childn >40 kg 4 tab as single dose for 3 consecutive days. Childn 31-40 kg 3 tab as single dose for 3 consecutive days, 21-30 kg 2 tab as single dose for 3 consecutive days, 11-20 kg 1 tab as single dose for 3 consecutive days.
Administration
Should be taken with food: Take w/ meals or a milky drink. Take at the same time each day.
Contraindications
Hypersensitivity. Severe renal impairment (CrCl <30 mL/min).
Special Precautions
Not for treatment of cerebral malaria or other severe manifestations of complicated malaria eg, hyperparasitaemia, pulmonary oedema or renal failure. Recrudescent infections due to P falciparum or failure of chemoprophylaxis; travellers w/ intense exposure to P vivax or P ovale & those who develop malaria caused by either of these parasites. Monitor parasitaemia in patients receiving metoclopramide or tetracycline. Not recommended w/ concomitant rifampicin or rifabutin. Severe renal impairment. Pregnancy. Not recommended during lactation. Paed patients <11 kg.
Adverse Reactions
Headache; abdominal pain, nausea, vomiting, diarrhoea. Anaemia, neutropenia; hyponatraemia, anorexia; insomnia, dizziness; elevated liver enzyme levels; rash; fever; cough.
Drug Interactions
Proguanil: Potentiated anticoagulant effect of warfarin & other coumarin-based anticoagulants. Atovaquone: Decreased plasma conc w/ tetracycline, metoclopramide, rifampicin & rifabutin. Decreased trough levels of indinavir.
