Prednisolone
Indications/Uses
Allergic, collagen, dermatological, GI, desquamative gingivitis, hematologic, tuberculous meningitis, ophth, resp, rheumatic & endocrine disorders; neoplastic disease; nephrotic syndrome; non-suppurative thyroiditis; hypercalcemia associated w/ cancer of sarcoidosis; trichinosis w/ neurological or myocardial involvement; prophylaxis of organ rejection.
Dosage/Direction for Use
Adult 5-60 mg/day as single or divided doses. Max: 250 mg/day. Childn 500 mcg-2 mg/kg or 15-60 mg/m2 daily in 3 divided doses. Adrenal Insufficiency Childn 140 mcg/kg or 4 mg/m2 daily in 3 divided doses.
Administration
Should be taken with food: Take immediately after meals.
Contraindications
Peptic ulcer, osteoporosis, psychoses & severe psychoneuroses. Acute infections including herpes zoster, herpes simplex, eye ulceration, TB except as adjunctive treatment. Vaccination w/ live vaccine.
Special Precautions
AIDS, cardiac disease, CHF, HTN, DM, ulcerative colitis, ocular herpes simplex, viral or bacterial infections, hyperlipidemia, hypothyroidism. Prolonged & high-dose therapy. Renal & hepatic dysfunction. May suppress growth & development in childn.
Adverse Reactions
Increased gluconeogenesis, hyperglycemia, electrolyte imbalance, oedema, corticotropin reaction inhibition, nitrogen depletion, spontaneous fracture, osteoporosis, posterior subcapsular cataract, Cushing's syndrome, amenorrhoea, hyperhidrosis, mental & neurological disturbances, intracranial HTN, thromboembolic complication, increased susceptibility to infections, acute pancreatitis, aseptic necrosis of bone, growth retardation in childn.
Drug Interactions
Increased ulcerogenic effect w/ alcohol or other anti-inflammatory drugs. Enhanced hypokalemia w/ amphotericin B, K-depleting diuretics. Increased or decreased effects w/ oral anticoagulants. Enhanced possibility of arrhythmias or digitalis toxicity w/ cardiac glycosides. Increased metabolic clearance w/ ephedrine, phenobarb, phenytoin or rifampicin. Potential GI ulceration & hemorrhage w/ heparin. Increased blood glucose conc of hypoglycemics. Salicylates, somatropin.
