Indications/Uses
Cryptococcosis; systemic, mucosal & genital candidiasis.
Dosage/Direction for Use
Cryptococcal meningitis & other infections 400 mg on 1st day followed by 200-400 mg once daily for 6-8 wk. Prevention of Cryptococcal meningitis relapse in patient w/ AIDS 200 mg daily. Candidemia, disseminated candidiasis & other invasive Candida infections 400 mg on 1st day followed by 200 mg daily. Prevention of candidiasis 50-400 mg once daily. Patient at high risk systemic infection 400 mg once daily. Systemic candidiasis & cyptococcal infections 6-12 mg/kg daily. Patient w/ renal impairment Initial dose: 50-400 mg. Adjust dose according to CrCl: >50 mL/min 100% recommended dose; ≤50 mL/min w/ no dialysis 50% recommended dose; regular dialysis 100% recommended dose after each dialysis.
Administration
May be taken with or without food.
Contraindications
Concomitant use w/ terfenadine & cisapride.
Special Precautions
Pregnancy & lactation.
Adverse Reactions
Headache, rash, abdominal pain, diarrhea, flatulence, nausea, hepatic toxicity, dizziness, seizures, alopecia, exfoliative skin disorders, dyspepsia, vomiting, leukopenia, hepatic failure, hypercholesterolemia, taste perversion.
Drug Interactions
Anticoagulants, benzodiazepines, cisapride, cyclosporin, hydrochlorothiazide, OC, phenytoin, rifabutin, rifampicin, sulfonylureas, tacrolimus, terfenadine, theophylline, zidovudine.
