Hyperuricemia associated w/ gout & gouty arthritis. Adjuvant to therapy w/ penicillin or ampicillin, methicillin, oxacillin, cloxacillin or nafcillin for plasma level elevation & prolongation.
Dosage/Direction for Use
Gout 250 mg bd for 1 wk, followed by 500 mg bd thereafter. May be increased by 0.5 g-increments every 4 wk w/in tolerance if symptoms are not controlled or 24-hr uric acid excretion is not >700 mg. Max: Not >2 g daily. Probenecid & penicillin therapy Adult 2 g daily in divided doses. Childn 2-14 yr Initially 25 mg/kg daily. Maintenance: 40 mg/kg daily in 4 divided doses.
Hypersensitivity. Known blood dyscrasia or uric kidney stones. Childn <2 yr.
Special Precautions
Hypersensitivity. Exacerbation of gout. Concomitant use w/ methotrexate, salicylates. Not recommended in conjunction w/ penicillin in known renal impairment. Women of childbearing potential. Pregnancy & lactation.
Adverse Reactions
Headache, GI symptoms, urinary frequency, hypersensitivity reactions eg, anaphylaxis, dermatitis, pruritus, fever, flushing, dizziness, anaemia.
Drug Interactions
Psychic disturbances w/ penicillin or other β-lactams. Antagonized uricosuric action by salicylates, pyrazinamide. Prolonged or enhanced action of oral sulfonylureas. Increased plasma conc of indomethacin, acetaminophen, naproxen, ketoprofen, meclofenamate, lorazepam, rifampin, methotrexate.
