Indications/Uses
Primary hypercholesterolaemia (type IIa including heterozygous familial hypercholesterolaemia), mixed dyslipidaemia (type IIb) or homozygous familial hypercholesterolaemia in patients not adequately responsive to diet & other appropriate measures. Prevention of CV events in patients at high risk of a 1st CV event.
Dosage/Direction for Use
Adult Hypercholesterolemia Initially 5-10 mg once daily, increased if necessary to 20 mg once daily for at least 4 wk interval. Severe hypercholesterolemia Increased further to 40 mg daily for 4 wk. Prevention of CV events 20 mg once daily. Elderly Hypercholesterolemia Initially, 5 mg once daily. Asian patients Initially 5 mg once daily. May be increased if necessary. Max: 20 mg daily. Concomitant use w/ fibrate: Initially 5 mg once daily. May be increased if necessary. Max: 20 mg daily.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Patients w/ active liver disease or w/ unexplained elevations in transaminase.
Adverse Reactions
Headache, myalgia, asthenia, constipation, dizziness & abdominal pain. Rare: Myositis, rhabdomyolysis, pancreatitis & hypersensitivity reaction.
Drug Interactions
Increased levels in concomitant use w/ itraconazole, fluconazole; 2-fold increased levels w/ gemfibrozil. Decreased levels w/ erythromycin, antacids containing Al & Mg hydroxide.
