Severe forms of cystic acne & acne conglobata, especially when the lesions involve the trunk. Correction of severe keratinization disorders.
Dosage/Direction for Use
Adult & adolescent 0.5-1 mg/kg in 2 divided doses daily for 15-20 wk. Maintenance: 0.1-1 mg/kg daily. Max: 2 mg/kg/day.
Should be taken with food.
Hypersensitivity. Hepatic or renal insufficiency. Hypervitaminosis A. Patients w/ excessively elevated blood lipid values. Blood donation during & w/in 1 mth of treatment to women of childbearing potential. Pregnancy & lactation.
Special Precautions
Diabetes, obesity, alcoholism & disorders of lipid metabolism. History of depression. Perform liver function tests & check serum lipids (fasting value) before & 1 mth after start of treatment & at 3 mth intervals thereafter. Concomitant use w/ contraceptives containing an androgen progestational substance. Dermabrasion & wax epilation. Increased risk of dental problems due to dry mouth. Avoid exposure to sun. Not indicated in concurrent radiation therapy w/ UV light. May cause vision impairment which may affect ability to drive or operate machinery.
Adverse Reactions
Chelitis, epistaxis & skin infection. Dryness of the skin, mucosa eg, lips, nasal mucosa (epistaxis), eyes (conjunctivitis, reversible corneal opacities & intolerance to contact lenses). Exanthema, pruritis, dermatitis facialis, sweating, pyogenic granuloma; paronychia, nail dystrophy, increased formation of granulation tissue, persistent hair thinning, reversible alopecia, acne fulminans, hirsutism, hyperpigmentation, photosensitivity; muscle pain, joint pain, hyperostosis, tendonitis; behavioral disorders, depression, increased intracranial pressure, seizures; isolated cases of visual disturbances, impaired hearing at certain frequencies, photophobia, decreased night vision, lenticular cataract, keratitis; nausea; inflammatory bowel disease eg, colitis, ileitis & hemorrhage. Transitory & reversible increase in transaminases; bronchospasm; decreased WBC count, RBC parameters, increase or decrease in platelet count, elevated sedimentation rate; increase in serum triglyceride & cholesterol levels, hyperuricemia; local or systemic infection due to gm +ve microorganisms; lymphadenopathy, hematuria & proteinuria, pancreatitis, vasculitis; bleeding or inflammation of gums; cataracts or corneal opacities.
Drug Interactions
Intensified symptoms of hypervitaminosis A w/ vit A. Pseudotumor cerebri w/ tetracyclines. May diminish effect of microdosed progesterone prep.
