Concise Prescribing Info
Metastatic colorectal cancer. Adjuvant treatment after surgery for Dukes C colon cancer. Locally advanced or metastatic breast cancer after failure of anthracycline-containing chemotherapy w/ docetaxel. After monotherapy & failure of taxanes & anthracycline-containing regimens for breast cancer & when anthracycline-containing therapy is not indicated. Advanced or metastatic breast cancer whose tumours overexpress human epidermal receptor type 2 (HER2) w/ lapatinib. Gastric cancer w/ platinum-based regimen.
Dosage/Direction for Use
Monotherapy in colon, colorectal, breast cancer 1.25 g/m2 bid for 14 days followed by 7-day rest period. Combination therapy in breast cancer w/ docetaxel Capecitabine: 1.25 g/m2 bid for 14 days followed by 7-day rest period. Docetaxel: 75 mg/m2 as  1 hr IV infusion every 3 wk. Combination therapy in colorectal or gastric cancer 0.8-1 g/m2 bid for 14 days, followed by 7-day rest period. Alternatively, 625 mg/m2 bid continuously.
Should be taken with food: Take 30 min after a meal. Swallow w/ water.
Special Precautions
Renal impairment increases systemic exposure to 5'-deoxy-5-fluorouridine.
Adverse Reactions
Diarrhoea, nausea, vomiting, abdominal pain, stomatitis, palmar-plantar erythrodysesthesia. Fatigue, asthenia & anorexia. Rashes, alopecia, erythema, dryness of the skin, pruritis, skin pigmentation disorders & nail disorders.
Drug Interactions
Altered coagulation parameters & bleeding w/ warfarin or phenprocoumon. Increased plasma conc of phenytoin. Fatal fluoropyrimidine toxicity w/ sorivudine or its analogues. Small increase in plasma conc w/ antacids containing Al or Mg. Reduced max tolerated dose w/ folinic acid or interferon-α.
