Olanzapine

Indications/Uses

Management of schizophrenia. Moderate to severe mania associated w/ bipolar disorder.

Dosage/Direction for Use

Schizophrenia Initially 10 mg daily as single dose at intervals not <24 hr to w/in the range of 5-20 mg daily. Treatment of acute mixed or manic episodes in bipolar disorder Initially 10-15 mg daily as monotherapy or 10 mg in combination therapy, may be adjusted in increments or decrements of 5 mg if necessary at intervals of not <24 hr to a dose between 5 & 20 mg daily. Prevention of recurrence in patient whose manic episodes have responded previously to olanzapine Initially 10 mg daily. Elderly (female) or non-smokers Initially 5 mg daily. Hepatic or renal impairment Initially 5 mg daily.

Administration

May be taken with or without food.

Contraindications

Angle-closure glaucoma.

Special Precautions

Cerebrovascular disease or conditions predisposing to hypotension; benign prostatic hyperplasia or paralytic ileus; Parkinson's disease; hepatic impairment, history of blood dyscrasias, bone marrow depression or myeloproliferative disease. Pregnancy & lactation.

Adverse Reactions

Hyperprolactinemia. Increased appetite, dizziness; elevated plasma glucose, triglyceride & liver enzyme values; eosinophilia, edema, orthostatic hypotension & mild transient antimuscarinic effects eg, constipation & dry mouth.
