Pregabalin
Indications/Uses
Adjunct in the treatment of partial seizures w/ or w/o secondary generalization. Treatment of generalized anxiety disorder.
Dosage/Direction for Use
150-600 mg/day given in 2 divided doses. Epilepsy Adult Initially 150 mg/day, may be increased to 300 mg/day after 1 wk. Max: 600 mg/day after an additional 7-day interval. Patient w/ renal impairment CrCl ≥60 mL/min 150-600 mg bid or tid. ≥30 to <60 mL/min 75-300 mg bid or tid. ≥15 to <30 mL/min 25-150 mg bid or qid. <15 mL/min 25-75 mg once a day. Supplementary dosage following haemodialysis 25-100 mg as single dose.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Galactose intolerance, Lapp lactase deficiency or glucose-galactose malabsorption. Angioedema.
Special Precautions
Hypersensitivity including angioedema. Increased risk of suicidal thoughts or behavior. Loss of vision, visual blurring or other changes of visual acuity. W/drawal symptoms. Renal failure. CHF. May impair ability to drive or operate machinery. Pregnancy & lactation. Childn <12 yr & adolescents 12-17 yr. Elderly >65 yr.
Adverse Reactions
Dizziness, somnolence. Increased appetite; euphoric mood, confusion, irritability, decreased libido, disorientation, insomnia; ataxia, abnormal coordination, tremor, dysarthria, memory impairment, disturbance in attention, paresthesia, sedation, balance disorder, lethargy, headache; blurred vision, diplopia; vertigo; vomiting, dry mouth, constipation, flatulence; urinary incontinence, dysuria; erectile dysfunction; abnormal gait, feeling drunk, fatigue, peripheral edema; increased wt.
Drug Interactions
May potentiate the effects of ethanol & lorazepam. Risk of resp failure & coma w/ other CNS depressants. Additive in the impairment of cognitive & gross motor function caused by oxycodone. Risk of reduced lower GI tract function w/ medications that have the potential to produce constipation eg, opioid analgesics.
