Quetiapine fumarate
Indications/Uses
Management of manifestations of psychotic disorders. Schizophrenia.
Dosage/Direction for Use
Initially 25 mg bid on day 1, 50 mg bid on day 2, 100 mg bid on day 3 & 150 mg bid on day 4. May be adjusted to 300-450 mg daily in 2 divided doses, although 150 mg daily may be adequate for some patient. Max: 750 mg daily. Elderly or impaired hepatic or renal function 25 mg daily, increased in steps of 25-50 mg daily.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Coma or severe CNS depression. Lactation.
Special Precautions
Perform eye examination to detect cataract before starting & every 6 mth during treatment. Pregnancy. Elderly. Childn.
Adverse Reactions
Extrapyramidal symptoms. Mild asthenia, anxiety, dizziness, myalgia, rhinitis, dyspepsia, increased plasma triglycerides & cholesterol conc, reduced plasma thyroid hormone conc; cataracts.
Drug Interactions
May enhance hypotensive effects w/ antihypertensives. Decreased oral clearance w/ cimetidine. Antagonism w/ dopamine agonists or levodopa. Increased mean oral clearance w/ phenytoin.
