Contents
Per tab Rifampicin 150 mg, INH 75 mg, pyrazinamide 400 mg, ethambutol HCl 275 mg
Indications/Uses
Treatment of both smear +ve pulmonary & extra-pulmonary TB, in adults in the intensive initial phase of treatment.
Dosage/Direction for Use
Adult >70 kg 5 tab/day; 55-70 kg 4 tab/day; 50 kg 3 tab/day. 38-54 kg 3 tab/day; 30-37 kg 2 tab/day. Duration: 4 mth.
Administration
Should be taken on an empty stomach: Take at least 30 min before meals.
Contraindications
INH-associated hepatic injury, optic neuritis, liver disease, hepatitis.
Special Precautions
Urine, feces, saliva, sputum, sweat & tears may be colored red-orange. Contact lens may be colored. Hypersensitivity, current chronic liver disease or severe renal dysfunction. Periodic ophth exam.
Adverse Reactions
GI disturbance, transient abnormalities in liver function tests, elevations in BUN & serum uric acid, anorexia, nausea, vomiting, abdominal pain, fever, malaise, headache. Rifampicin: Thrombocytopenia, leukopenia, hemolytic anemia, eosinophilia & decreased Hb, pruritus, urticaria, rash, pemphigoid reaction, sore mouth, sore tongue & exudative conjunctivitis. INH: Peripheral neuropathy, elevated serum transaminases (SGOT; SGPT), bilirubinemia, bilirubinuria, jaundice, fatal hepatitis. Agranulocytosis, hemolytic sideroblastic or aplastic anemia, thrombocytopenia & eosinophilia, fever, skin eruptions, lymphadenopathy, vasculitis, pyridoxine deficiency, pellagra, hyperglycemia, metabolic acidosis, gynecomastia, rheumatic syndrome, lupus erythematosus-like syndrome. Ethambutol: Optic neuritis, anaphylactoid reactions, dermatitis, pruritus, joint pain, dizziness, mental confusion, disorientation & hallucination. Numbness & tingling of the extremities. Pyrazinamide: Flushing, hepatitis, arthralgia, hyperuricemia, acute gouty arthritis.
Drug Interactions
Other anti-TB drugs, methasone, oral hypoglycemics, digitoxin, quinidine, disopyramide, dapsone, corticosteroid. Phenytoin, alcohol.
