Amoebiasis, balantidiasis & Blastocystis hominis infection, giardiasis, trichomoniasis, bacterial vaginosis, acute necrosing ulcerative gingivitis, prevention of post-op anaerobic bacterial infection, PUD, leg ulcers & pressure sores & anaerobic infections.
Dosage/Direction for Use
Tab Adult Amoebiasis, balantidiasis, Blastocystis hominis infection 400-800 mg tid for 5-10 days or 1.5-2.5 g as single dose daily for 2 or 3 days. Giardiasis 2 g once daily for 3 days or 400 mg tid for 5 days or 500 mg bid for 7-10 days. Trichomoniasis 2 g as single dose or 2-day course of 800 mg in the morning & 1.2 g in the evening or 7-day course of 600 mg to 1 g daily in 2 or 3 divided doses. Bacterial vaginosis 400-500 mg bid for 5-7 days or 2 g as single dose. Acute necrosing ulcerative gingivitis 200-250 mg every 8 hr for 3 days. Prevention of post-op anaerobic bacterial infections 400 mg every 8 hr, 24 hr before surgery followed post-op by infusion or rectally until oral therapy is possible. PUD 400 mg bid (except when combined w/ omeprazole & amoxicillin) or 400 mg tid for 1 wk. Leg ulcers & pressure sores 400 mg every 8 hr for 7 days. Anaerobic infections Initially 800 mg then 400 mg every 8 hr for 7 days or 500 mg every 8 hr. Susp Childn Trichomoniasis 7-day course: 3-7 yr 100 mg tid or 15 mg/kg/day in divided doses, 1-3 yr 50 mg tid. Giardiasis & amoebiasis 15 mg/kg in divided doses; 7-10 yr 35-50 mg/kg daily in divided doses. Anaerobic infections 7.5 mg/kg/day every 8 hr.
Should be taken on an empty stomach: Take at least 1 hr before meals.
Hypersensitivity to imidazoles. Monitor leukocyte count.
Special Precautions
Active or chronic severe peripheral & CNS diseases. Severe hepatic impairment. Alcoholic beverages. Avoid unnecessary use. Pregnancy & lactation.
Adverse Reactions
GI disturbances mostly nausea & unpleasant metallic taste. Diarrhea, constipation & vomiting. Furred or coated tongue, glossitis, stomatitis. Headache, weakness, dizziness, drowsiness, ataxia, dysarthria, insomnia, changes in mood or mental state eg, depression & confusion, seizure & peripheral neuropathy. Temporary moderate leucopenia, thrombocytopenia, skin rashes, urticaria, pruritus. Optic neuropathies, urethral discomfort, darkening of urine, elevated liver enzymes, cholestatic hepatitis & jaundice.
Drug Interactions
May impair metabolism or excretion of warfarin, phenytoin, lithium, ciclosporin & fluorouracil. Increased metabolism w/ phenytoin. Decreased plasma conc w/ phenobarb. Increased plasma conc w/ cimetidine. May provoke disulfiram-like reactions w/ alcohol.
