Indications/Uses
Treatment & prophylaxis of hemorrhage associated w/ excessive fibrinolysis. Prophylaxis of hereditary angioedema. Inj General fibrinolysis, as in prostatic & pancreatic cancer, after thoracic & other major surgery. Obstet complications eg, abruption placentae & post-partum haemorrhage. Leukemia & liver diseases in connection w/ thrombolytic therapy w/ streptokinase. Hereditary angioneurotic edema.
Dosage/Direction for Use
Cap 1-1.5 g (15-25 mg/kg) bid-qid. Inj 5-10 mL by slow IV inj at a rate of 1 mL/min bid-tid. Prostatectomy 5-10 mL by slow IV inj every 8 hr (1st inj given during operation) for the 1st 3 days after surgery. Epistaxis Applied topically by soaking a gauze strip in the soln then packing the nasal cavity. Dental surgery in patients w/ hemophilia 10 mg/kg IV immediately before surgery. General fibrinolysis 10 mL by slow IV inj tid-qid. Renal impairment CrCl >500 μmol/L 5 mg/kg daily, 250-100 μmol/L 10 mg/kg daily, 120-250 μmol/L 10 mg/kg bid.
Administration
May be taken with or without food.
Contraindications
Inj: Hypersensitivity. Active thromboembolic disease eg, DVT, pulmonary embolism & cerebral thrombosis. Subarachnoid hemorrhage. Patient w/ acquired defective color vision.
Special Precautions
Patients w/ active IV clotting, predisposition to thrombosis, hemorrhage due to disseminated intravascular coagulation. Inj: Irregular menstrual bleeding. Renal impairment. Hematuria from the upper urinary tract. Regularly perform eye exam during long-term use. Pregnancy & lactation.
Adverse Reactions
Dose-dependent GI disorders (nausea, vomiting, diarrhea). Dizziness &/or hypotension; thrombotic complications.
Drug Interactions
Inj: Avoid blood transfusion or inj containing penicillin.
