Indications/Uses
To reduce LDL-cholesterol, apolipoprotein B & triglycerides, & to increase HDL-cholesterol in the treatment of hyperlipidemias including hypercholesterolemias & combined (mixed) hyperlipidemia (type IIa or IIb hyperlipoproteinemias), hypertriglyceridemia (type IV) & dysbetalipoproteinemia (type III). Adjunctive therapy in patients w/ homozygous familial hypercholesterolemia who have some LDL-receptor function.
Dosage/Direction for Use
10-20 mg once daily. Patient requiring large reduction in LDL-cholesterol Initially 40 mg daily. May adjust dose at intervals of 4 wk up to a max of 80 mg daily. Familial hypercholesterolemia Initially 10 mg daily then increased at intervals of at least 4 wk to 40 mg once daily. Max: 80 mg once daily. Childn & adolescent 10-17 yr Heterozygous familial hypercholesterolemia Initially 10 mg once daily. Max: 20 mg once daily.
Administration
May be taken with or without food: Avoid excessive consumption (>1 L/day) of grapefruit juice.
Contraindications
Special Precautions
Should not be given to patients w/ acute liver disease or unexplained persistently raised serum aminotransferase conc. Discontinue if marked or persistent increase in serum aminotransferase or creatinine phosphokinase conc occur. Asses liver function before initiation of treatment & monitor periodically until 1 yr after the last evaluation in dose. Severe renal impairment. Avoid during pregnancy.
Adverse Reactions
GI disturbances. Headache, skin rashes, dizziness, blurred vision, insomnia & dysgeusia. Reversible increase in serum aminotransferase conc.
