Indications/Uses
To reduce LDL-C, Apo B & triglycerides, & to increase HDL-C in the treatment of hyperlipidemias including hypercholesterolemias & combined (mixed) hyperlipidemia (type IIa or IIb hyperlipoproteinemias). Adjunct therapy in patients w/ HoFH who have some LDL-receptor function. Prophylaxis to hypercholesterolemic patients w/ ischemic heart disease.
Dosage/Direction for Use
Initially 5-10 mg in the evening. Ischemic heart disease Initially 20 mg, may be adjusted at intervals of not <4 wk up to a max of 80 mg once daily in the evening. HoFH 40 mg once daily in the evening or 80 mg daily in 3 divided doses of 20 mg, 20 mg, & an evening dose of 40 mg. Patient taking cyclosporine, fibric acid derivatives or nicotinic acid & risk of myopathy Max: 10 mg daily.
Administration
May be taken with or without food: Avoid excessive consumption (>1 L/day) of grapefruit juice.
Special Precautions
Should not be given to patients w/ acute liver disease or unexplained persistently raised serum aminotransferase conc. Discontinue if marked or persistent increases in serum aminotransferase or creatine phosphokinase conc occur. Severe renal impairment. Avoid during pregnancy.
Adverse Reactions
GI disturbances. Headache, skin rashes, dizziness, blurred vision, insomnia, dysgeusia.
Drug Interactions
Risk of myopathy w/ cyclosporine, fibric acid derivatives, or nicotinic acid.
