Sildenafil citrate
Indications/Uses
Management of erectile dysfunction.
Dosage/Direction for Use
Adult 50 mg 1 hr before sexual activity. Max: 100 mg once daily. Impaired renal & hepatic function 25 mg.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Concomitant use w/ organic nitrates. Pregnancy & lactation.
Special Precautions
Consider CV status of the patient prior to initiating therapy. Patients w/ anatomical deformation of the penis eg, angulation, cavernal fibrosis or Peyronie's disease, or conditions which may predispose to priapism. Co-administration w/ α-blocker. Patients w/ bleeding disorders or active peptic ulceration; retinitis pigmentosa.
Adverse Reactions
Headache, flushing, dyspepsia. Dizziness, vomiting, diarrhea, hypotension, tachycardia, syncope, epistaxis, nasal congestion, rash, eye pain, red eyes/bloodshot eyes, muscle/back pain, prolonged erection &/or priapism. UTI, cerebrovascular hemorrhage, transient ischemic attacks.
Drug Interactions
Reduced clearance w/ CYP3A4 inhibitors eg, ketoconazole, erythromycin, cimetidine. Increased plasma conc w/ cimetidine. Increased systemic exposure w/ erythromycin. Increased Cmax & AUC w/ saquinavir & ritonavir. Potentiate hypotensive effects of nitrates. Symptomatic hypotension w/ α-blocker therapy.
