Acyclovir

Indications/Uses

200 mg: HSV infections, genital herpes of the skin & mucous membranes (ie, primary & frequently recurring genital herpes). Recurrence prevention for adults w/ very severe forms of very frequently recurring genital HSV infection. 400 mg: Prevention of severe cases of herpes simplex infection in severely immunocompromised adults exposed to an elevated risk of infection (eg, following organ transplants). 400 mg & 800 mg: Shingles (herpes zoster).

Dosage/Direction for Use

200 mg HSV infections Adult & childn >2 yr 1 tab 5 times daily (every 4 hr) during the day. Childn <2 yr ½ adult dose. Patient w/ renal impairment (CrCl <10 mL/min/1.73 m²) 1 tab bd (every 12 hr). Prophylaxis of severe forms & very frequently recurring genital HSV infections Immunocompetent adult patient 1 tab qds (every 6 hr); or 1 tab tds (every 8 hr) or 1 tab bd (every 12 hr). Immunocompromised adult patient 1 tab qds (every 6 hr). 200 mg & 400 mg Prophylaxis of severe forms & very frequently recurring genital HSV infections or HSV infection prevention Severely immunocompromised patient (eg, following organ transplant) 400 mg (two 200-mg tab or one 400-mg tab) qds (every 6 hr). 400 mg & 800 mg Shingles Adult 800 mg (two 400-mg tab or one 800-mg tab) 5 times daily (every 4 hr) during the day. Patient w/ renal impairment: CrCl 10-25 mL/min/1.73 m² 800 mg tds (every 8 hr), <10 mL/min/1.73 m² 800 mg bd (every 12 hr).

Overdosage

View Acyclovir 200/400/800 Stada overdosage for action to be taken in the event of an overdose.

Administration

Should be taken with food: Take after meals. Swallow whole w/ a glass of water. Ensure adequate fluid intake.

Contraindications

Hypersensitivity. Renal impairment; reduced urinary output <100 mL/24 hr (for 200 mg & 400 mg only).

Special Precautions

Pregnancy. Breastfeeding is discouraged. Check kidney function & ensure adequate fluid intake among elderly.

Adverse Reactions

Skin rash; GI symptoms including nausea, vomiting, diarrhoea & abdominal pain; dizziness, confusion, hallucinations, drowsiness; transient convulsions & psychoses.

Drug Interactions

Reduced renal excretion by probenecid.
