Amphotericin B
Indications/Uses
Potentially life-threatening fungal infections.
Dosage/Direction for Use
0.1 mg/mL by slow IV infusion over 2-6 hr. Initially 0.25 mg/kg body wt daily over 2-6 hr. Patient w/ good cardiopulmonary function, tolerance & w/o severe reaction Initially 0.3 mg/kg body wt daily. Patient w/ impaired cardiopulmonary function or severe reaction Initially smaller dose of 5-10 mg daily. May be increased gradually by 5-10 mg/day to 0.5-0.7 mg/kg body wt/day. Max: 1.5 mg/kg/day. Treatment duration: 6-12 wk or longer.
Contraindications
Hypersensitivity. Hepatic disease.
Special Precautions
Monitor liver & renal function, serum electrolytes (especially Mg & K) & blood counts frequently.
Adverse Reactions
Fever, malaise, wt loss, GI disturbances, hematologic disorders, muscle & joint pains, headache, decreased & abnormal renal function.
Drug Interactions
Cisplatin, pentamidine, aminoglycosides, cyclosporine, corticosteroids, corticotropin, digitalis glycoside, muscle relaxants & antiarrhythmic agents.
