Zidovudine
Indications/Uses
HIV-1 infection in combination with other antiretrovirals. Prevention of maternal-fetal HIV transmission.
Dosage/Direction for Use
Adult & childn >12 yr Combination therapy w/ other antiretrovirals 600 mg daily in divided doses (200 mg every 8 hr or 300 mg every 12 hr). Childn 3 mth-12 yr 180 mg/m2 body surface every 6 hr. Max: 200 mg every 6 hr. Prevention of maternal-fetal HIV transmission Pregnant HIV-infected women 100 mg 5 times a day initiated at 14-34 wk of pregnancy & continued until the start of labor. When labor begins, IV zidovudine therapy should be substituted. Neonate 2 mg/kg every 6 hr, initiated w/in 12 hr after birth & continued through 6 wk.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Significant anemia or granulocytopenia.
Special Precautions
Hepatic or renal disease. Elderly. Perform blood cell count & indices of anemia regularly.
Adverse Reactions
Bone marrow depression, neutropenia. Nausea, vomiting, abdominal pain, anorexia, headache, insomnia, myalgia, fever, rash.
Drug Interactions
Paracetamol may increase the incidence of neutropenia. Other drugs metabolized by hepatic glucuronidation, nephrotoxic or bone marrow suppressants, probenecid, ribavirin.
