Indications/Uses
Effective treatment in the following infections caused by susceptible anaerobic bacteria; susceptible strains of gm +ve aerobic bacteria eg, streptococci, staphylococci & pneumococci & susceptible strains of Chlamydia trachomatis. Upper & lower resp infections; specific skin & soft tissue infections; diabetic foot infection, gynecological infections; intra-abdominal infections; dental infections; septicemia & endocarditis; toxoplasmic encephalitis & Pneumocystis carinii pneumonia in patients w/ AIDS; malaria. Prophylaxis of endocarditis in patients who are sensitive/allergic to penicillins.
Dosage/Direction for Use
Adult 150-300 mg every 6 hr. Childn >1 mth 8-20 mg/kg/day in 3-4 divided doses. β-hemolytic streptococcal infections 150-300 mg every 6 hr for at least 10 days. Diabetic foot infections Mild/moderate: 300 mg qid for 7-14 days. Chlamydia trachomatis cervicitis 450 qid for 14 days. Toxoplasmic encephalitis in patients w/ AIDS 600 mg 6 hrly w/ oral pyrimethamine (200 mg loading dose followed by 50-75 mg once daily) & oral leucovorin (10-20 mg once daily) continued for at least 6 wk. Pneumocystis carinii pneumonia in patients w/ AIDS 300-450 mg 6 hrly & primaquine 15-30 mg once daily for 21 days. Acute streptococcal tonsillitis/pharyngitis 300 mg bid for 10 days. Malaria Adult 20 mg/kg daily in 3 equally divided doses for 7 days in conjunction w/ quinine sulfate 650 mg tid for 3-7 days. Childn 20 mg/kg daily in 3 equally divided doses for 7 days in conjunction w/ quinine sulfate 10 mg/kg tid for 3-7 days. Prophylaxis of endocarditis in patients sensitive to penicillins Adult 600 mg 1 hr before procedure. Childn 20 mg/kg 1 hr before procedure.
Administration
May be taken with or without food: Swallow whole w/ a full glass of water & in an upright position.
Contraindications
Hypersensitivity.
Special Precautions
Discontinue if mild cases of pseudomembranous colitis occur. Patients w/ diarrhea subsequent to the administration of antibacterial agents. Should not be used in the treatment of meningitis. In prolonged therapy, liver & kidney function test must be performed. Pregnancy & lactation.
Adverse Reactions
Transient neutropenia & eosinophilia; anaphylactoid reactions; dysgeusia; thrombophlebitis; abdominal pain, nausea, vomiting & diarrhea, esophagitis & esophageal ulcer; jaundice; maculopapular rash & urticaria.
Drug Interactions
Erythromycin, neuromuscular blocking agents.
