Glibenclamide
Indications/Uses
Type 2 DM.
Dosage/Direction for Use
Initially 2.5 mg daily increasing by 2.5 mg at 1-2 wk intervals until optimal metabolic control is achieved. Max daily dose: 15 mg. Max single dose: 10 mg.
Administration
Should be taken with food: Take just before breakfast or a main meal, & take any remaining portion of the total daily dose just before the evening meal.
Contraindications
Type I DM, diabetic coma, diabetic ketoacidosis, severe renal impairment & hepatic dysfunction. Patients treated w/ bosentan. Pregnancy & lactation.
Special Precautions
Cross-sensitivity to sulfonamides & sulfonamide derivatives.
Adverse Reactions
GI disturbances. Hypersensitivity reactions. Blood dyscrasias.
Drug Interactions
Bosentan. Potentiation of hypoglycemic effect w/ insulin & other (oral) antidiabetics, ACE inhibitors, anabolic steroid & male sex hormones, azapropazone, chloramphenicol, coumarin derivatives, cyclophosphamide, disopyramide, fenfluramine, fenyramidol, fibrates, fluoxetine, ifosfamide, MAOIs, miconazole, oxyphenbutazone, para-aminosalicylic acid, pentoxifylline, phenylbutazone, probenecid, quinolones, salicylates, sulfinpyrazone, sulphonamides, sympatholytic agents, clarithromycin, tetracycline, tritoqualine, trofosfamide. Weakening of the blood sugar-lowering effect by acetazolamide, barbiturates, diazoxide, diuretics, epinephrine & other sympathomimetic agents, glucagon, corticosteroids, laxatives, nicotinic acid, oestrogens & progestogens, phenothiazines, phenytoin, thyroid hormones, rifampicin.
