Indications/Uses
In combination w/ 5-FU & folinic acid (LV): Adjuvant treatment of stage III (Duke's C) colon cancer after complete resection of primary tumor, metastatic colorectal cancer, unresectable advanced or metastatic gastric cancer & unresectable hepatocellular carcinoma.
Dosage/Direction for Use
Adjuvant treatment of colon cancer 85 mg/m2 IV in combination w/ 5-FU/LV base regimen repeated every 2 wk for 12 cycles. Metastatic colorectal cancer 85 mg/m2 IV in combination w/ 5-FU/LV base regimen repeated every 2 wk. Unresectable advanced or metastatic gastric cancer 85 or 100 mg/m2 IV in combination w/ 5-FU/LV base regimen repeated every 2 wk or 130 mg/m2 IV in combination w/ epirubicin & capecitabine or 5-FU repeated every 3 wk. Unresectable hepatocellular carcinoma 85 mg/m2 IV in combination w/ 5-FU/LV base regimen repeated every 2 wk.
Contraindications
Lactation. Hypersensitivity. Bone marrow depression & peripheral sensory neuropathy w/ functional impairment prior to treatment.
Special Precautions
Neurological exam should be performed before each administration is started & then periodically. For patients who develop acute laryngopharyngeal dysaesthesias, during or w/in hr following the 2-hr infusion, the next oxaliplatin infusion should be administered over 6 hr. Monitor blood counts. If hematological toxicity occurs (neutrophils <1,500/mm3 or platelet counts <50,000/mm3), administration of the next course of therapy should be postponed until toxicity has resolved.
Adverse Reactions
Anemia, leucopenia, Grade 3-4 thrombocytopenia; nausea, vomiting & diarrhea; sensory peripheral neuropathy, characterized by dysaesthesia &/or paraesthesia in the extremities, neurological toxicity, fever & rash.
