Adjunctive therapy in the treatment of partial seizures w/ & w/o secondary generalization in patients >12 yr w/ epilepsy & in childn 3-12 yr. Management of postherpetic neuralgia in adults.
Dosage/Direction for Use
Epilepsy Adult & childn >12 yr Usual dose: 900-1,800 mg/day given in divided doses tid. Initially 300 mg tid. Dose may be increased using 300 mg tid up to 1,800 mg/day. Childn Initially 10-15 mg/kg/day in 3 divided doses & effective dose reached upward titration over a period of approx 3 days, 5-12 yr 25-35 mg/kg/day given in 3 divided doses, 3-12 yr Dose up to 50 mg/kg/day, 3-4 yr 40 mg/kg/day given in 3 divided doses. Postherpetic neuralgia Adult & childn >12 yr Usual dose: 900 mg/day given as 3 equally divided doses, may be increased based on response. Max: 3,600 mg/day. Initially 300 mg once daily on day 1, bid on day 2 & tid on day 3. Dose can be subsequently titrated up to 1,800 mg daily (divided tid).
May be taken with or without food: Swallow whole w/ sufficient fluid.
Special Precautions
Increased risk of suicidal thoughts or behavior. Monitor for emergence or worsening of depression, suicidal thoughts or behavior &/or any unusual changes in mood or behavior. Do not abruptly discontinue. Liver & kidney disease. Hematologic disorder. May affect ability to drive or operate machinery. Pregnancy & lactation.
Adverse Reactions
Dizziness, somnolence, ataxia, fatigue; viral infection.
Drug Interactions
Reduced bioavailability w/ antacids. Increased AUC w/ morphine.
