Indications/Uses
Management of type 2 DM (NIDDM) as an adjunct to diet & exercise to lower blood glucose. In combination w/ metformin or insulin in patients whose hyperglycemia cannot be controlled by diet & exercise in conjunction w/ a single oral hypoglycemic agent.
Dosage/Direction for Use
Initially 1-2 mg once daily. Maintenance dose: 1-4 mg once daily, may be increased in 2-mg increments at 1-2 wk intervals after a dose of 2 mg once daily. Max: 8 mg once daily. In combination w/ insulin Initially 8 mg once daily.
Administration
Should be taken with food: Take immediately before or during breakfast, or the 1st main meal of the day. Do not skip meals.
Contraindications
Hypersensitivity to glimepiride or sulfonamides. Diabetic ketoacidosis.
Special Precautions
Severe hypoglycemia. Renal or hepatic impairment. Malnourishment. Regularly check glucose levels in blood & urine, & HbA1c. Patients w/ G6PD. Rare hereditary problems of galactose intolerance. Pregnancy & lactation. Childn. Elderly.
Adverse Reactions
Dizziness, headache, hypoglycemia, nausea & weakness.
Drug Interactions
Potentiation of blood sugar-lowering effect w/ insulin & other oral antidiabetics, ACE inhibitors, allopurinol, anabolic steroids & male sex hormones, chloramphenicol, coumarin derivatives, cyclophosphamide, disopyramide, fenfluramine, fenyramidol, fibrates, fluoxetine, guanethidine, ifosfamide, MAOIs, miconazole, para-aminosalicylic acid, quinolones, salicylates, sulfinpyrazone, sulfonamides, tetracyclines, tritoqualine, trofosfamide. Raised blood sugar level w/ acetazolamide, barbiturates, corticosteroids, diazoxide, diuretics, epinephrine & other sympathomimetic agents, glucagons, laxatives, nicotinic acid (high dose), estrogens & progestogens, phenothiazines, phenytoin, rifampicin, thyroid hormones.
