Amoxicillin trihydrate
Indications/Uses
Infections caused by susceptible gm -ve bacteria (Haemophilus influenzae, E. coli, Proteus mirabilis, Salmonella) & gm +ve bacteria (Strep pneumoniae, enterococci, non-penicillinase-producing Staphylococci, Listeria) eg, otitis media, resp tract infection, ear, nose & throat, GIT, GUT, skin & soft tissues.
Dosage/Direction for Use
Ear, nose, throat, skin, & GUT infections Adult & childn over 12 yr 500 mg every 12 hr or 250 mg every 8 hr. Dose may be increased to 500 mg every 8 hr in severe infection. Duration of therapy: Continue for at least 2-3 days after patient becomes asymptomatic. Gonorrhea Single 3 g oral dose. Prevention of bacterial endocarditis Single 2 g oral dose given 1 hr before dental procedure or minor upper resp tract surgery or instrumentation.
Administration
May be taken with or without food: May be taken w/ meals for better absorption & to reduce GI discomfort.
Contraindications
Special Precautions
History of allergy, impaired renal function. Periodic determination of renal, hepatic & hemopoietic function during prolonged therapy is recommended. Discontinue use if skin rashes, itching or edema, flu-like syndrome, Stevens-Johnson syndrome, blistering, peeling skin & conjunctivitis. May cause anaphylactic reactions. Pregnancy & lactation.
Adverse Reactions
Agitation, anxiety, confusion, dizziness, headache, insomnia, seizure; erythematous maculopapular rash, Stevens-Johnson syndrome, urticaria; nausea, vomiting, diarrhea; agranulocytosis, anemia; increased ALT & AST.
Drug Interactions
Synergistic effect against β-lactamase producing bacteria w/ clavulanic acid. May decrease renal secretion of methotrexate. Increase serum results in GI or hematologic toxicity of methotrexate. Slowed rate of renal secretion w/ oral probenecid (if administered shortly before amoxicillin). Potentiated aminopenicillin allergy may occur w/ allopurinol.
