Oxaliplatin
Indications/Uses
Metastatic colorectal cancers after failure of treatment w/ fluoropyrimidines, alone by monochemotherapy or along w/ fluoropyrimidines.
Dosage/Direction for Use
130 mg/m2 IV over at least 2 hr, every 3-4 wk. In combination w/ 5-FU, w/ or w/o folinic acid: 130 mg/m2 every 3 wk or 85 mg/m2 every 2 wk.
Contraindications
Severe preexisting peripheral neuropathy, severe renal dysfunction (CrCl <30 mL/min). Pregnancy & lactation.
Special Precautions
Paraesthesia or functional impairment (discontinue).
Adverse Reactions
Sensory neuropathy, anemia, fever, nausea & vomiting, liver function abnormalities, infections, alopecia, pharyngolaryngeal dysesthesia. In combination w/ 5-FU & leucovorin: Neutropenia, thrombocytopenia, diarrhea, mucositis.
Drug Interactions
Potentiates irinotecan inhibition of acetylcholinesterase. Avoid Al-containing IV administration sets & alkaline soln.
