Sildenafil citrate
Indications/Uses
Erectile dysfunction.
Dosage/Direction for Use
Adult 50 mg 30 min to 4 hr before sexual activity. Max 100 mg once daily. Elderly >65 yr, patients w/ renal (CrCl <30 mL/min) & hepatic impairment, on stable α-blockers & potent CYP3A4 inhibitors Initially 25 mg. W/ ritonavir Max: 25 mg 48 hrly.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity; concurrent use of organic nitrates in any form (eg, nitroglycerin, isosorbide dinitrate).
Special Precautions
Visual disturbance, hearing loss, hypotension, vision loss, priapism & prolonged erection. Anatomical penis deformation, bleeding disorders, CV diseases, sickle cell anemia, multiple myeloma, leukemia, PUD, hepatic & renal impairment.
Adverse Reactions
Flushing, headache, insomnia, paresthesia, dizziness, erythema, rash, epistaxis, rhinitis, abnormal vision, dyspepsia, diarrhea, gastritis, UTI, myalgia, dyspnea, sinusitis, pyrexia.
Drug Interactions
α-blockers (may cause symptomatic hypotension). Increased level w/ CYP3A4 inhibitors (eg, ritonavir). Nitrate (only if 24 hr elapsed). Other drugs for erectile dysfunction.
