Ofloxacin
Indications/Uses
Resp tract, ophth, GUT, GIT, skin & soft tissue infections & diseases. Otitis media, UTI; bones & joints infections; septicaemia; multidrug resistant TB.
Dosage/Direction for Use
Adult 200-600 mg daily in 1-3 divided doses. Uncomplicated gonorrhoea 400 mg single dose. 2nd-line anti-TB drug in treatment of apparent or proved multidrug resistant TB 600-800 mg daily given at 12-hr intervals.
Administration
May be taken with or without food: Avoid antacids or supplements containing Fe or Zn w/in 2 hr before or after ofloxacin. Ensure adequate hydration.
Contraindications
Hypersensitivity to ofloxacin & other quinolones.
Special Precautions
Severe renal disorders, seizure or history of convulsion, myasthenia gravis. Pregnancy & lactation. Childn. Elderly.
Adverse Reactions
Shock, toxic epidermal necrolysis, mucocutaneous ocular syndrome (Stevens-Johnson syndrome), allergic reactions, QT prolongation, ventricular tachycardia, acute renal failure, interstitial nephritis, fulminant hepatitis, jaundice, transient elevation of SGOT, SGPT, alkaline phosphatase, γ-GTP, total bilirubin, GI symptoms, transient decrease in leukocytes, erythrocytes, Hb, hematocrit, platelets & increase in eosinophils, agranulocytosis, pancytopenia, thrombocytopenia & hemolytic anemia, insomnia, dizziness, headache, psychoneurologic disorders, interstitial & eosinophilic pneumonia, rhabdomyolysis, hypoglycemia, tendon disorders, psychiatric symptoms, hypersensitivity vasculitis, exacerbation of myasthenia gravis.
Drug Interactions
Antacids containing Al or Mg & drugs containing Fe may reduce absorption. NSAIDs of phenylacetic/propionic acid derivatives may cause convulsion. May enhance effect of warfarin.
