Indications/Uses
Dosage/Direction for Use
Adult & adolescent ≥12 yr Duodenal ulcer 20 mg once daily for 2-4 wk. Severe cases: may be increased to 40 mg once daily for 4 wk. Relapse prophylaxis of duodenal & gastric ulcer 20-40 mg/day. H. pylori eradication Triple therapy: 20 mg, amoxicillin 1 g & clarithromycin 500 mg bd for 1 wk or 20 mg, clarithromycin 250 mg & metronidazole 500 mg (or tinidazole 500 mg) bd for 1 wk or 40 mg daily, amoxicillin 500 mg & metronidazole 500 mg tds for 1 wk. Gastric ulcer & reflux esophagitis 20 mg once daily for 4-8 wk. Severe cases: may be increased to 40 mg once daily for 8 wk. NSAID-associated gastroduodenal ulcers or erosions w/ or w/o concomitant NSAID therapy 20 mg once daily for 4-8 wk. Max: 40 mg. Prevention of relapse in NSAID-associated peptic ulcers, gastroduodenal erosions & dyspeptic symptoms 20 mg once daily. Childn ≥20 kg body wt 20-40 mg/day; >5 mth, <20 kg body wt 1-1.4 mg/kg/day; ≤5 mth 0.5-1 mg/kg/day, for 4 wk. Max: 8 wk. Symptomatic GERD 20 mg once daily for 4 wk. Zollinger-Ellison syndrome Initially 60 mg once daily. Severe cases: May be increased to 80-120 mg daily in 2 divided doses.
Administration
May be taken with or without food.
Special Precautions
GI malignancy. May affect ability to drive or operate machinery. Pregnancy & lactation.
Adverse Reactions
Headache, diarrhoea, constipation, abdominal pain, nausea, vomiting, flatulence.
Drug Interactions
Ketoconazole, itraconazole. May delay elimination of diazepam & phenytoin. Clarithromycin.
