May be taken with or without food.
Contraindications
Special Precautions
Menstruation is suppressed during treatment. Increased incidence of endometrial changes including hyperplasia, polyps, cancer & uterine sarcoma. 2nd primary tumours. Any patients who report abnormal gynaecological symptoms, especially vag bleeding or who presents w/ menstrual irregularities. McCune Albright syndrome. In delayed microsurgical breast reconstruction, it may increase the risk microvascular flap complications. Venous thromboembolism. Very rarely, radiation recall in patients who received prior radiotherapy. Women should not become pregnant. Lactation. Childn.
Adverse Reactions
Nausea; fluid retention; vag bleeding, discharge; skin rash; hot flushes; fatigue. Anaemia; cataracts, retinopathy; hypersensitivity reactions; elevated triglycerides; leg cramp, myalgia; uterine fibroids; ischaemic cerebrovascular events, headache, light headedness, sensory disturbances (including paraesthesia & dysgeusia); pruritus valvae, endometrial changes (including hyperplasia & polyps); alopecia; vomiting, diarrhoea, constipation; changes in liver enzymes, fatty liver; thromboembolic events (including DVT, microvascular thrombosis & pulmonary embolism).
Drug Interactions
Increased anticoagulant effect w/ coumarin-type anticoagulants. Increased risk of thromboembolic events w/ cytotoxic agents. Reduced levels w/ rifampicin (CYP3A4 inducer), CYP2D6 inhibitors.
