Pregabalin
Indications/Uses
Peripheral neuropathic pain in adults. Adjunctive therapy in adults w/ partial seizures w/ or w/o secondary generalization. Relieve generalized anxiety disorder & pain in fibromyalgia.
Dosage/Direction for Use
150-600 mg daily in 2 or 3 divided doses. Neuropathic pain Initially 75 mg twice daily, may be increased to 150 mg twice daily after 3-7 days, & further to a max of 300 mg twice daily after an additional wk if needed. Epilepsy Initially 75 mg twice daily, may be increased to 150 mg twice daily after 1 wk, & further to a max of 300 mg twice daily after an additional wk if needed. Generalized anxiety disorder 150-600 mg daily in 2-3 divided doses. Initially 150 mg daily & may be increased to 300 mg daily after 1 wk, then it can be further increased to 450 mg daily on the next following wk. Max dose: 600 mg daily in the additional wk. Fibromyalgia 300-450 mg daily in 2 divided doses. Initial dose: 75 mg twice daily, may be increased to 150 mg twice daily after 1 wk, then it can be further increased to 225 mg twice daily on the next following wk. Patient w/ renal impairment CrCl ≥60 mL/min 150 mg daily in 2 divided doses. Max: 600 mg daily. CrCl 30-60 mL/min 75 mg daily in 1 or 2 divided doses. Max: 300 mg daily. CrCl 15-30 mL/min 25-50 mg daily in 1 or 2 divided doses. Max: 150 mg daily. CrCl <15 mL/min 25 mg single dose daily. Max: 75 mg daily. Supplementary dose following haemodialysis 25 mg single dose. Max: 100 mg.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity.
Special Precautions
Not to be used in patients w/ rare hereditary problems of galactose intolerance, the Lapp lactase deficiency or glucose-galactose malabsorption. Hypersensitivity reactions can occur, including cases of angioedema. Blurred vision. CHF. Elderly >65 yr. May increase occurrence of accidental injury (fall) in the elderly.
Adverse Reactions
Dizziness, somnolence. Increased appetite; euphoric mood, confusion, decreased libido, irritability; ataxia, disturbance in attention, abnormal coordination, memory impairment, tremor, dysarthria, paresthesia; blurred vision, diplopia; vertigo; dry mouth, constipation, vomiting, flatulence; erectile dysfunction; fatigue, peripheral edema; feeling drunk, edema, abnormal gait; increased wt.
Drug Interactions
Additive effects in the impairment of cognitive & gross motor function w/ oxycodone. May potentiate the effects of ethanol & lorazepam.
