Domperidone
Indications/Uses
Dyspeptic symptom complex associated w/ delayed gastric emptying eg, epigastric sense of fullness, abdominal distension or swelling, epigastric pain or discomfort. Acute symptoms of nausea & vomiting. May be suitable for chemotherapy- or radiotherapy-induced nasuea & vomiting or post-op nausea & vomiting.
Dosage/Direction for Use
Adult Chronic dyspepsia 1 tab 3 times daily. Nausea & vomiting 1 tab 3 times daily. Max: 30 mg daily. Childn Chemotherapy- or radiotherapy-induced nausea & vomiting 0.25 mL/kg 3-4 times daily.
Administration
Should be taken on an empty stomach: Take 15-30 min before meals.
Contraindications
Hypersensitivity. Prolactin-releasing pituitary tumour (prolactinoma). GI haemorrhage, mechanical obstruction or perforation. Co-administration w/ potent CYP3A4 inhibitors eg, azole antifungals (itraconazole, voriconazole, posaconazole), macrolide antibiotics (clarithromycin, telithromycin), PIs (ritonavir, saquinavir, telaprevir). Moderate or severe hepatic impairment.
Special Precautions
Patients w/ current or history of cardiac disease. Increased risk of serious ventricular arrhythmias & sudden cardiac death. Co-administration w/ potent CYP3A4 inhibitors which have not been shown to cause QT interval prolongation eg, indinavir. Monitor closely for signs & symptoms of AR.
Drug Interactions
Anti-dyspeptic effect may be antagonized by anticholinergic drugs. Reduced relaxant effect of atropine if administered first. Increased plasma levels & QT prolongation w/ CYP3A4 inhibitors. May influence absorption of concomitantly administered drugs eg, SR or EC formulations due to its gastrokinetic effects.
