Sildenafil citrate
Indications/Uses
Dosage/Direction for Use
Adult 50 mg approx 1 hr before sexual activity. Max: 100 mg once daily. Hepatic & severe renal impairment 25 mg once daily. Concomitant therapy w/ ritonavir Max: 25 mg as single dose for 48-hr period. Concomitant therapy w/ CYP3A4 inhibitors eg, erythromycin, saquinavir, ketoconazole, itraconazole Initially 25 mg.
Administration
May be taken with or without food.
Contraindications
Hypersensitivity. Concurrent use w/ nitric oxide, organic nitrates or nitrites.
Special Precautions
Not to be used in men for whom sexual activity is inadvisable. Co-administration w/ α-blocker & other agents for erectile dysfunction. Left ventricular outflow obstruction, anatomical penis deformation or conditions predisposing to priapism, bleeding disorders or active peptic ulceration, retinitis pigmentosa.
Adverse Reactions
Headache; vasodilatation. Dizziness; abnormal vision, chromatopsia; palpitation; rhinitis; dyspepsia.
Drug Interactions
Reduced clearance w/ CYP3A4 inhibitors (eg, ketoconazole, erythromycin, cimetidine). Increased Cmax & AUC w/ ritonavir. Symptomatic postural hypotension w/ doxazosin.
