Granisetron HCl

Indications/Uses

Prevention & treatment of acute & delayed nausea & vomiting associated w/ chemo- & RT (CINV & RINV); post-op nausea & vomiting (PONV).

Dosage/Direction for Use

Adult CINV 1-3 mg slow IV inj over 30 sec or IV infusion over 5 min prior to chemotherapy. Max: 9 mg/24 hr. RINV prevention 1-3 mg slow IV inj over 30 sec or as IV infusion over 5 min prior to RT. PONV prevention 1 mg slow IV inj over 30 sec prior to anesth induction. PONV treatment 1 mg slow IV inj over 30 sec. Max: 3 mg daily. Childn CINV 10-40 mcg/kg IV infusion over 5 min prior to chemotherapy, may administer additional dose w/in 24 hr at least 10 min after initial infusion.

Special Precautions

Signs of subacute intestinal obstruction; preexisting arrhythmias or cardiac conduction disorders; cardiac comorbidities. Patients on cardiotoxic chemotherapy &/or w/ concomitant electrolyte abnormalities. Cross-sensitivity w/ other 5-HT₃ antagonists. Concomitant use w/ other serotonergic drugs. Pregnancy & lactation.

Adverse Reactions

Headache; constipation. Elevated hepatic transaminases.

Drug Interactions

Increased total plasma clearance w/ phenobarb. Serotonin syndrome w/ other serotonergic drugs. Concurrent use w/ drugs prolonging QT interval &/or are arrhythmogenic.
